Hybridization probe design

Now it is possible to let QuantPrime design hybridization probes - either together with primer pairs (e.g. for TaqMan assays) or alone (e.g. for QISH - Quantitative In-Situ Hybridization).

This feature is configured in custom primer design parameter sets - on the "Create new project" page, select "Custom" as quantification protocol and create your own primer pair design and specificity testing parameters.

A few options to optimize probe selection (especially important for TaqMan assays) are configurable; avoidance of 5' G's and selection of the strand with most C's. It is also possible to force the strand on which the probe is designed - necessary for applications that involves direct hybridization with mRNA, like QISH.

We are open for further suggestions to improve these new features, and to add related features, such as Tm calculations for probes with added minor groove binders and/or partially containing LNA. We would be delighted to hear about your experimental experience with primers and probes designed with QuantPrime.
